PRINCIPLE

The surfactants present in the reagent release indirect bilirubin allowing it to react, together with the conjugate, with the diazonium salt (DPD) forming a red azobilirubin compound in an acid medium. The intensity of the colour produced in the reaction is directly proportional to the concentration of total bilirubin present in the sample.

DIAGNOSTIC USE

Bilirubin is a breakdown product of hemoglobin that is transported to the liver together with albumin. There it is conjugated with glucuronic acid and is secreted through bile. This conjugated bilirubin is the direct bilirubin, while unconjugated bilirubin is called indirect bilirubin. Total bilirubin in serum is equivalent to direct bilirubin plus indirect bilirubin. High level of indirect or total bilirubin may indicate liver disorders, biliary obstructions, hemolytic diseases, physiological neonatal jaundice, Gilbert’s disease or fructose intolerance.
