PRINCIPLE

At pH 2.5 the formation of the complex between the pyrogallol red – molybdate and the proteins produces a change in the absorbance at 600 nm. This change is proportional to the protein concentration and can be measured spectrophotometrically.

DIAGNOSTIC USE

Protein concentration measures commonly CSF-brain barrier integrity. Higher concentrations are associated with inflammatory processes. These values are increased in bacterial meningitis, tuberculosis, brain abscess, stroke, encephalomyelitis, and other degenerative processes that cause neurological disease. High protein concentrations are found in urine when there is renal injury, such as nephrotic syndrome, glomerulonephritis, renal failure and malignant renal tumors. A single test result could not be used to make a clinical diagnosis. It should integrate clinical and laboratory data.
